Abstract

Marine brown algae are well-recognized repositories of structurally diverse secondary metabolites with considerable pharmacological potential. The present study investigated the qualitative phytochemical composition and quantitative phenolic and flavonoid contents of petroleum ether, ethanol, and aqueous extracts of two ecologically important brown algae — Sargassum wightii Greville and Padina pavonica (L.) Thivy — freshly collected from the intertidal zone of Mandapam, Ramanathapuram District, Tamil Nadu, India. Shade-dried powdered material was subjected to sequential cold maceration in three solvents of ascending polarity, and the resulting crude extracts were screened for ten classes of secondary metabolites using standard colorimetric and precipitation methods (Harborne, 1998; Trease and Evans, 2002). Total phenolic content (TPC) and total flavonoid content (TFC) were determined spectrophotometrically by the Folin–Ciocalteu and aluminium chloride methods respectively, with all assays performed in triplicate. Sargassum wightii yielded the most chemically diverse profile: alkaloids, cardiac glycosides, phenols, flavonoids, tannins, steroids, terpenoids, proteins, and carbohydrates were detected, with the ethanol extract recording the highest TPC (18.42 ± 0.94 mg GAE/g dw) and TFC (11.63 ± 0.82 mg QE/g dw). Padina pavonica contained flavonoids, tannins, saponins, steroids, terpenoids, alkaloids, phenols, proteins, and carbohydrates across the three solvents, with ethanol again yielding the highest TPC (8.36 ± 0.62 mg GAE/g dw) and TFC (2.87 ± 0.31 mg QE/g dw). The distribution of compound classes across solvents of ascending polarity demonstrates that extraction solvent is a primary determinant of metabolite recovery. These findings establish the chemical foundation for subsequent bioactivity-focused studies on both species.
